Backfire thru the carb?
Timing is off or sticking intake valve. Spark is igniting while an intake valve is open.
I think you should not depend on one backfire and check that it is getting spark every time.
Sounds like timing is off, or is it old enough that it uses points? If so, put in a new set of points, and gap correctly.

My 460 cranks like a champ and has spark at the plugs. Every couple cranks, it backfires out the carb (one of the reasons I know it has spark). The gas was bad, and with starting fluid, it didn't even try to start. I tried fresh gas, and starting fluid. Nothing. Any ideas?
